Abstract

Moringa oleifera Lam., commonly known as the drumstick tree, is a medicinal plant widely recognized for its rich nutritional profile and diverse therapeutic properties. It contains a variety of phytoconstituents including flavonoids, phenolic compounds, alkaloids, tannins, saponins, and terpenoids that contribute to its pharmacological activities. The present study was undertaken to standardize and evaluate the phytochemical constituents of Moringa oleifera leaves using pharmacognostic, physicochemical, phytochemical, and chromatographic techniques. Physicochemical parameters such as ash values, extractive values, moisture content, crude fiber, and foreign organic matter were determined according to standard pharmacopoeial procedures. Preliminary phytochemical screening was carried out using n-hexane and 70% ethanolic extracts to identify the major classes of secondary metabolites. Quantitative estimation of total phenolic content, total flavonoid content, and tannin content were performed using spectrophotometric methods. Antioxidant activity was evaluated by the DPPH free radical scavenging assay, while Thin Layer Chromatography (TLC) and High-Performance Thin Layer Chromatography (HPTLC) were employed for chromatographic fingerprinting and quantification of marker flavonoids. The ethanolic extract demonstrated higher concentrations of phenolic compounds and flavonoids along with significant antioxidant activity compared to the n-hexane extract. HPTLC fingerprint analysis confirmed the presence of characteristic flavonoid constituents, supporting the quality and authenticity of the plant material. The findings indicate that Moringa oleifera possesses a rich phytochemical profile with considerable antioxidant potential and highlight the importance of standardization for ensuring the quality, safety, and therapeutic efficacy of herbal medicines.
